The interaction between conserved viral late domains and the cellular ESCRT proteins (NEDD4, TSG101 and Alix) represents a crucial step in the replication/egress of several viruses, making these PPIs promising but underexplored targets for the development of novel Broad-Spectrum Antivirals (BSAs). We report herein our ongoing efforts to obtain TSG101-targeting antivirals, offering a promising opportunity to identify broad-spectrum agents that are less susceptible to resistance development.
