Redraspberries,containingellagitanninsandcyanidin-basedanthocyanins,werefedtovolunteersand metabolites appearinginplasmaandurinewereanalysedbyUHPLC-MS.Anthocyaninswerenotab- sorbed toanyextentwithsubnmol/Lconcentrationsofcyanidin-3-O-glucoside andacyanidin-O-glucuronide appearingtransientlyinplasma.Anthocyaninsexcretedinurinecorrespondedto0.007%of intake. MoresubstantialamountsofphaseIImetabolitesofferulicacidandisoferulicacid,alongwith4′- hydroxyhippuricacid,potentiallyoriginatingfrompH-mediateddegradationofcyanidinintheproximal gastrointestinaltract,appearedinurineandalsoplasmawherepeakconcentrationswereattained1– 1.5hafterraspberryintake.Excretionof18anthocyanin-derivedmetabolitescorrespondedto15.0%of intake, a figure substantiallyhigherthanobtainedinotheranthocyaninfeedingstudies.Ellagitannins pass fromthesmalltothelargeintestinewherethecolonicmicrobiotamediatetheirconversionto urolithins AandBwhichappearedinplasmaandwereexcretedalmostexclusivelyassulfateandglu- curonide metabolites.Theurolithinmetabolitespersistedinthecirculatorysystemandwereexcretedin urine formuchlongerperiodsoftimethantheanthocyaninmetabolitesalthoughtheiroverallurinary recoverywaslowerat7.0%ofintake.Itiseventsoriginatingintheproximalanddistalgastrointestinal tract, andsubsequentphaseIImetabolism,thatplayanimportantroleinthebioavailabilityofboth anthocyaninsandellagitanninsanditistheirmetaboliteswhichappearinthecirculatorysystem,that are keytoelucidatingthemodeofaction(s)underlyingtheprotectiveeffectsofthesecompoundson human health.
