The aim of this work was the evaluation of mucoadhesion using LiveBox2 cell from IVTech (Pisa, Italy), a dynamic millifluidic platform, and porcine esophageal epithelium as model for buccal mucosa. As reference, inclined plane technique, rheological analysis and tack-test were performed. The formulation tested was a polymeric gel Carbopol 974P NF, loaded with lidocaine as model drug. In particular, the effect of polymer concentration on the measured parameters was studied. From the experimental data, an increasing mucoadhesive property of the formulations was observed as the concentration of Carbopol 974P NF increased. Both the traditional methods and the innovative single dynamic model showed a linear correlation (R2>0,95) between mucoadhesion and the polymer content, verifying the sensitivity and suitability of the developed system. A linear correlation (R2>0.95) between the parameters measured with the IVTech cells and the other methods was found highlighting the good capability of the innovative single dynamic technique to predict mucoadhesion.
